X-ray micro-computed tomography (CT) produces three-dimensional images of samples on a micrometer scale. This technique has several advantages, such as its nondestructive character and low measurement time, compared with other techniques. However, when applied to biological samples of soft tissue, the low attenuation and low effective contrast between structures pose difficulties in creating appropriate images for morphological studies. Diffusible iodine-based contrast-enhanced CT (DICE-CT), which uses iodine solutions to enhance contrast, is a viable alternative for addressing the aforementioned challenges. Given the variety of biological samples, an appropriate methodology must be adapted depending on the dimensions and morphological characteristics of the investigated object. A specimen that has not been morphologically studied by micro-CT and is of high sanitary importance is the adult Aedes aegypti mosquito. This study investigated the stage of iodine staining in the treatment of the A. aegypti mosquito to determine the most suitable staining time for the morphological study of this mosquito in adulthood. After determining the appropriate staining time, we discuss the potential of applying DICE-CT and methodology to mosquito studies. Seven A. aegypti females were treated using fixation steps with Bouin's solution, dehydration in a graded ethanol series, staining with iodine solution (1%), and washing in absolute ethanol. Only the staining step was different between samples. Each mosquito spent a varying amount of time (6-72 h) in the iodine solution (1%). For comparison, one of the mosquitoes was not stained. After treatment, the samples were scanned using the Bruker SkyScan 1172 micro-CT scanner. The reconstructed volumes and histograms were compared to determine the most suitable time. In addition, a quantitative analysis was performed based on a comparison of the attenuation profiles of the mosquito brains. Thereafter, the most suitable treatment process was selected, and two other samples were scanned after applying the selected process. Although fewer than 18 h was insufficient for an effective increase in attenuation and effective contrast, surpassing 24 h proved unnecessary and resulted in saturating the gray tones visualized through the histograms, leading to information loss. Therefore, a time of approximately 24 h was the most suitable staining time for studying adult A. aegypti. It was possible to isolate the organs of the digestive and reproductive systems of the mosquito stained for 24 h. Thus, micro-CT was confirmed to be an excellent technique in studies of individual structures of adult A. aegypti mosquitoes.

Background: Lipoxin A4 (LXA4) has anti-inflammatory and pro-resolutive roles in inflammation. We evaluated the effects and mechanisms of action of LXA4 in titanium dioxide (TiO2) arthritis, a model of prosthesis-induced joint inflammation and pain. Methods: Mice were stimulated with TiO2 (3mg) in the knee joint followed by LXA4 (0.1, 1, or 10ng/animal) or vehicle (ethanol 3.2% in saline) administration. Pain-like behavior, inflammation, and dosages were performed to assess the effects of LXA4 in vivo. Results: LXA4 reduced mechanical and thermal hyperalgesia, histopathological damage, edema, and recruitment of leukocytes without liver, kidney, or stomach toxicity. LXA4 reduced leukocyte migration and modulated cytokine production. These effects were explained by reduced nuclear factor kappa B (NFκB) activation in recruited macrophages. LXA4 improved antioxidant parameters [reduced glutathione (GSH) and 2,2-azino-bis 3-ethylbenzothiazoline-6-sulfonate (ABTS) levels, nuclear factor erythroid 2-related factor 2 (Nrf2) mRNA and Nrf2 protein expression], reducing reactive oxygen species (ROS) fluorescent detection induced by TiO2 in synovial fluid leukocytes. We observed an increase of lipoxin receptor (ALX/FPR2) in transient receptor potential cation channel subfamily V member 1 (TRPV1)+ DRG nociceptive neurons upon TiO2 inflammation. LXA4 reduced TiO2-induced TRPV1 mRNA expression and protein detection, as well TRPV1 co-staining with p-NFκB, indicating reduction of neuronal activation. LXA4 down-modulated neuronal activation and response to capsaicin (a TRPV1 agonist) and AITC [a transient receptor potential ankyrin 1 (TRPA1) agonist] of DRG neurons. Conclusion: LXA4 might target recruited leukocytes and primary afferent nociceptive neurons to exert analgesic and anti-inflammatory activities in a model resembling what is observed in patients with prosthesis inflammation.

The objective of the study was to evaluate the circulating levels of chemical elements after exercise-induced muscle damage (EIMD) followed by the repeated bout effect (RBE). Seven physically active subjects (26.5 ± 4.0 years) performed two sessions of EIMD (5 sets of 20 drop jumps), the second session 14 days after the first for RBE assessment. Blood collections, countermovement jump (CMJ), squat jump (SJ), and delayed-onset muscle soreness (DOMS) were performed before (Pre), after (Post), and 24, 48, and 72 h after the exercise session. Creatine kinase (CK) was detected by biochemical analysis and the concentration of chemical elements by total reflection X-ray fluorescence (TXRF). Differences between time points and sessions were assessed with two-way ANOVA and the effect size (ES). EIMD induced a reduction in the CMJ at 24 h (P < 0.05) and an increase in DOMS at 24 h (P < 0.01) and 48 h (P < 0.01), and CK at 72 h (P < 0.05). RBE alleviated all symptoms of EIMD in the second session (P > 0.05). EIMD induced a large to very large ES for Zn reduction at 24 h (- 1.37) and 72 h (- 0.93) and Br (- 0.83) at 72 h. RBE presented large to very large ES for the increase in P at 48 h (0.92); Cl at 24 h (1.04); K at 24 h (0.91), 48 h (1.10), and 72 h (0.96); Ca at 72 h (0.92); and Fe at 24 h (0.85). RBE influenced the concentration of elements associated with fatigue (K, Ca, Cl), inflammatory response, and glucose metabolism (Zn).

The saliva analysis by Total Reflection X-Ray Fluorescence (TXRF) for monitoring training loads could be useful due to the ease of sample analysis and a large range of chemical elements can be detected. Twelve BJJ athletes were submitted to 7 weeks of training scheduled with high training loads (weeks 1 to 4) and tapering (weeks 5 to 7). Saliva samples were collected before (Pre), at week 4, and at week 8 (Post), to quantify chemical element concentrations by TXRF. The internal training load was monitored using the session rating of perceived exertion of session (RPE) method for physical and technical tactical training sessions. All the saliva samples presented Phosphorus (P), Sulfur (S), Chlorine (Cl), Potassium (K), Calcium (Ca), Zinc (Zn), Bromine (Br), and Rubidium (Rb) at Pre, 4 weeks and Post time points. Salivary concentrations of Cl, K, Manganese (Mn), Copper (Cu), Br, and Rb significantly decreased in 4 weeks (p<0.05); and K, Cu, and Rb concentration remained at low levels at Post (p<0.05). Medium to large Cohen´s effect sizes for decrements in element concentrations from Pre to 4 weeks was observed for all the elements except Zn. Reduced S concentration at 4 weeks was negatively correlated to high training load period (r=-0.56, p=0.05). The results suggest that BJJ training modulates salivary electrolyte composition and salivary P, S and Rb are correlated with training loads in BJJ athletes. (AU)


A análise de saliva por meio de Fluorescência de Raios-X de Reflexão Total (TXRF) para monitorar cargas de treinamento pode ser útil devido à facilidade de análise da amostra e a ampla variedade de elementos químicos detectados. Doze atletas de BJJ foram submetidos a 7 semanas de treinamento programado com altas cargas de treinamento (semanas 1 a 4) e polimento (semanas 5 a 7). Amostras de saliva foram coletadas antes (Pré), na semana 4 (Meio) e na semana 8 (Pós), para quantificar as concentrações dos elementos químicos por TXRF. A carga interna de treinamento foi monitorada por meio do método de percepção subjetiva de esforço da sessão (PSE) nas sessões de treinamento físico e técnico-tático. Todas as amostras de saliva apresentaram Fósforo (P), Enxofre (S), Cloro (Cl), Potássio (K), Cálcio (Ca), Zinco (Zn), Bromo (Br) e Rubídio (Rb) no momento Pré, 4 semanas e Pós. As concentrações salivares de Cl, K, Manganês (Mn), Cobre (Cu), Br e Rb diminuíram significativamente em 4 semanas (p <0,05); e a concentração de K, Cu e Rb permaneceram em níveis baixos no Pós (p <0,05). Os tamanhos de efeito de Cohen médio a grande para decréscimos nas concentrações de elementos, de Pré a 4 semanas, foram observados para todos os elementos, exceto para Zn. A redução da concentração de S em 4 semanas foi negativamente correlacionada com o período de alta carga de treinamento (r = -0,56, p = 0,05). Os resultados sugerem que o treinamento de BJJ modula a composição eletrolítica salivar e o P, S e Rb salivares estão correlacionados com as cargas de treinamento no BJJ. (AU)

BACKGROUND: Silver diamine fluoride has attracted attention because of its clinical success in arresting dental caries. Thus, the aim of this study was to evaluate the effect of different application times and concentrations of silver diamine fluoride (SDF) on deciduous tooth enamel remineralization. MATERIAL AND METHODS: Blocks of deciduous tooth enamel were categorized into six groups of 11 each: 2 control groups: intact enamel, and demineralized enamel; 38% SDF and 30% SDF which were subdivided according to application times (1 and 3 min). The microhardness of samples was determined, and all groups except the intact enamel group were subjected to pH cycling to produce initial carious lesions. The 38% and 30% SDF solutions were applied to the enamel for 1 or 3 min. After pH cycling and SDF treatments, the microhardness was again determined. Samples were sectioned to evaluate the cross-sectional microhardness. Furthermore, internal porosity of the samples was examined using micro-CT. Data were statistically analyzed by analysis of variance followed by Tukey's test, and linear regression analyses were performed. RESULTS: There was no difference in enamel remineralization based on surface and cross-sectional microhardness. The 30% SDF solution applied for 3 min promoted significantly less pores than the other groups. CONCLUSIONS: The 1-min application time promoted enamel remineralization regardless of the SDF concentration (30% or 38%). This work aimed to evaluate the salivary concentration of chemical elements in patients undergoing orthodontic treatment with fixed orthodontic appliances and removable aligners. Twelve Angle Class I and II orthodontic patients undergoing treatment with conventional fixed appliances and 15 patients treated with removable aligners provided unstimulated whole saliva samples before treatment (pre) and after 3 months of treatment (post). The concentration and secretion rate of chemical elements in saliva were determined by total reflection X-ray fluorescence. Differences from pre to post and between groups were determined with the paired T test or Wilcoxon test, and two-way ANOVA, considering P < 0.05. The concentrations of S, Cl, and K decreased, while Zn increased significantly (P < 0.05) between pre and post treatment with the fixed appliance treatment. The salivary secretion rate of S was decreased from pre to post in the fixed appliance group. No differences in the concentration and secretion rate of chemical elements were detected from pre to post in the Invisalign group. Fe secretion rate presented an interaction between time and treatment, with lower secretion at post (P = 0.02) in the Invisalign group. Increased Br secretion rate and decreased Rb, Fe, P, and K in Invisalign patients suggested a better salivary electrolyte profile regarding periodontal bone remodeling. No significant alterations in ions associated with metal corrosion and inflammatory reactions were detected in orthodontic patients under dental plaque control.

The aim of this study was to monitor the circulating and salivary ion concentrations by total reflection X-ray fluorescence (TXRF) in futsal players submitted to the futsal-specific intermittent shuttle protocol (FISP). TXRF may allow identification of changes in ion concentrations induced by physical efforts. Saliva and blood samples of 13 male futsal players were collected before (Pre) and after (Post) the FISP. Salivary and plasma ion levels were detected by TXRF, and differences from Pre to Post (paired t test or Wilcoxon test) and correlations between both biological fluids were determined (P < 0.05). All saliva samples presented phosphorus (P), sulfur (S), chlorine (Cl), potassium (K), calcium (Ca), iron (Fe), zinc (Zn), bromine (Br), and rubidium (Rb). S, Cl, Ca, Fe, Cu, Zn, Br, and Rb were detected in all blood samples. K, Cu, Br, and Rb presented reduced secretion rate from Pre to Post samples (P < 0.05). The salivary concentrations of K (r = - 0.53) and Zn (r = 0.54) were correlated with plasmatic concentrations. After FISP, salivary secretion of S (r = - 0.76), Cl (r = - 0.64), P (r = - 0.67), Mn (r = - 0.74), and Zn (r = 0.69) were correlated with plasma levels. We concluded that TXRF may be used to monitor salivary (P, S, Cl, K, Ca, Fe, Zn, Br, and Rb) and circulating (S, Cl, Ca, Fe, Cu, Zn, Br, and Rb) levels of several elements in futsal athletes. However, an acute bout of futsal-specific physical effort did not significantly imbalance ion concentrations in saliva or plasma.

INTRODUCTION: This in vitro study compared the performance of the XP-endo Shaper (XP; FKG, La Chaux-de-Fonds, Switzerland) in curved canals when used with a higher speed (3000 rpm) without a glide path with the manufacturer's protocol. METHODS: Twenty extracted mandibular molars with separate mesial curved canals were matched to obtain 2 standardized groups (n = 20). For the XP 1000 group, a glide path up to a size 15 hand file was performed followed by rotary instrumentation with the XP shaper at 1000 rpm, following the manufacturer's recommendations. For the canals in the XP 3000 group, the file was rotated at 3000 rpm after only negotiation the canal with a size 8 hand file (patency file). The operating time and the number of strokes taken to reach the working length (WL) and fit a 30/.04 gutta-percha cone to the WL were recorded. The shaping abilities were evaluated by micro-computed tomographic imaging and file deformation by scanning electron microscopy. Data were analyzed using analysis of variance and chi-square tests at a 5% significance level. RESULTS: The XP 3000 group required less time and strokes to reach the WL (P < .05) and resulted in a higher percentage of adequate cone fit (P < .05). No difference was found between groups regarding the micro-CT parameters, except for the taper, which was larger in the XP 3000 group in the apical and middle thirds (P < .05). No difference was found regarding file deformation between the 2 groups. CONCLUSIONS: The suggested protocol for the use of the XP (3000 rpm without a glide path) appears to be more efficient than the manufacturer's recommended protocol to prepare curved canals in vitro.

Aedes aegypti and Aedes albopictus are vectors of several arboviruses responsible for causing dengue, chikungunya, Zika, and yellow fever viruses and microcephaly, resulting in a public health problem in several countries worldwide. Even in this scenario, studies on the morphology of mosquito eggs are still lacking. In this paper, X-ray microtomography was used to study mosquito egg morphology. Several parameters such as length, surface area, volume, area-by-volume ratio, eccentricity and center of mass of the eggs were determined. The results obtained showed that micro-CT is an efficient technique for the morphological study of insect eggs and provides information that cannot be obtained with other techniques.

OBJECTIVE: To investigate the effect of two light-curing protocols on mechanical behavior of three bulk-fill resin composites (BFRC) considering their optical properties. METHODS: One increment of 4 mm thickness of the bulk-fill resin composites Opus Bulk Fill, Tetric N-Ceram and Filtek Bulk Fill Flow were submitted to two different light-curing protocols: Sp - irradiance of 1000 mW/cm2 (20 s); Xp - irradiance of 3200 mW/cm2 (6 s). To assess the influence on the mechanical behavior it was studied polymerization shrinkage by X-ray microtomography (n = 3), Vickers hardness (n = 10) at the top and bottom surfaces of the samples, irradiance reaching the bottom surface (n = 3) and absorbance spectrum during the light-curing time interval (n = 3). Data were analyzed by two-way ANOVA test for parametric data and Kruskal Wallis test, followed by Wilcoxon or Mann-Whitney U post-test, for non-parametric data. RESULTS: All BFRCs contracted when light-cured, with greater contraction for Xp. Filltek Bulk Fill Flow showed highest polymerization shrinkage, for both Sp and Xp. All BFRCs showed minor hardness values on the bottom surface, with greater reduction for Xp. All BFRCs exhibited a decrease in irradiance at 4 mm depth. A decrease in absorbance intensity throughout the light-cure was observed, except for Opus Bulk Fill. CONCLUSIONS: Regardless BFRCs composition, the light-curing protocol with lower irradiance and longer exposure time results in lower polymerization shrinkage and higher hardness. The higher irradiance in a shorter time interval compromises the mechanical behavior of the resin composites, which may result in undesirable clinical outcomes.

INTRODUCTION: This study evaluated the accuracy of 3 electronic apex locators (EALs) (CanalPro [Coltene-Endo, Cuyahoga Falls, OH], Apex ID [SybronEndo, Glendora, CA], and Root ZX Mini [J Morita, Tokyo, Japan]) in curved mesial canals of extracted mandibular molars using micro-computed tomographic (micro-CT) scanning. METHODS: The root canal length and the actual working length of 58 canals were measured using the visual method and 3-dimensional micro-CT reconstructions. The measurements of the EALs at marks "APEX/0.0" and "0.5 mark" were recorded as the electronic root canal length and the electronic working length, respectively. The absolute mean values and the percentages of distribution of the electronic measurements were compared with the actual lengths of the canals. The presence of isthmuses, accessory canals, foramen position, and the distance between the apical constriction and the apical foramen were recorded from the micro-CT scans, and their correlation to the accuracy of the EALs was determined. RESULTS: All electronic measurements showed high agreement with their respective gold standard, except the electronic root canal length of the Apex ID (P < .05). No difference in the percentage of precise measurements (within the ±0.5 mm) was found among the devices (χ2 and z tests, P > .05). Of the anatomic parameters evaluated, the presence of a lateral foramen negatively affected the 0.0 mark of the Apex ID (Wilcoxon test, P < .05). CONCLUSIONS: The Root ZX Mini and CanalPro were precise for both root canal length and working length determination in mesial curved canals of mandibular molars, whereas the Apex ID was accurate for the working length when using the 0.5 mark.

Previous studies have suggested that phototherapy may modulate orthodontic tooth movement and the incidence of root resorption. We aimed to identify a minimal dose-response relationship to LED therapy with regard to orthodontic tooth movement (OTM) and root resorption in rats. Forty-eight male Wistar rats were divided into six groups with equal and random distribution: control (C) no intervention; three daily LED irradiation (CLED); submitted only to OTM (RR); OTM and LED irradiation on the first day (LED1); OTM and two LED irradiation on the first and second days (LED2); and OTM and three LED irradiation on the first, second, and third days (LED3). Orthodontic appliance was installed in groups RR, LED1, LED2, and LED3 to promote OTM. Animals from groups CLED, LED1, LED2, and LED3 received LED therapy (940 nm, 4 J, 4 J/cm2) according to each group of treatment. After 7 days, all the animals were sacrificed. The jaws were fixed and scanned with microtomography (micro-CT). The micro-CT images were reconstructed on 2D and 3D models. These models were used to identify and measure root resorption number and dimensions (diameter, depth, and volume). The distance between the first and second molars was used to verify tooth displacement. The results showed that LED3 group had significantly lower number of root resorption. The root resorption dimensions (diameter and depth) had no significant differences among the experimental groups. LED3 group had significant tooth displacement in relation to C and CLED groups. In conclusion, three daily LED therapy doses are required to inhibit root resorption after appliance of orthodontic forces.

A study was carried out to evaluate the association of levels of radioactivity, selenium and aflatoxin in shelled Brazil nuts, which were classified in different sizes, for export. The selenium determinations were performed by inductively coupled plasma optical emission spectrometry (LOQ = 3.0 µg g-1), and aflatoxins were detected by Liquid chromatography-mass spectrometry (LOQ = 0.85 µg kg-1), recovery rates were between 92 and 100 percent. Radioactivity was measured by high-resolution gamma spectrometry. The selenium mean concentration was (22.7 ± 7.4) µg g-1. (n = 30). Mean activities determined for the following radium isotopes were: 15.77 Bq kg-1 for 224Ra, 104.8 Bq kg-1 for 226Ra and 99.48 Bq kg-1 for 228Ra. For 226Ra, the levels did not vary significantly with nut sizes, although such differences were observed for 224Ra and 228Ra. There was no statistically significant association between the level of selenium and the activity of radionuclides, however, there was correlation between the radionuclides. Aflatoxins above the quantification limit were not found.


Um estudo foi realizado para avaliar a associação dos níveis de radioatividade, selênio e aflatoxinas em castanha-do-Brasil descascada, que foram classificadas em diferentes tamanhos, para exportação. As determinações de selênio foram realizadas por espectrometria de emissão óptica com plasma indutivamente acoplado (LOQ=3,0 µg kg-1) e aflatoxinas foram detectadas por LC-MSMS (LOQ=0,85 µg kg-1), as taxas de recuperação ficaram entre 92 e 100 por cento. A radioatividade foi medida por espectrometria gama de alta resolução. A concentração média de selênio foi de (22,7 ± 7,4) µg g-1. (n = 30). A atividade média determinada para os radioisótopos foram: 15,77 Bq kg-1 para o 224Ra, 104,8 Bq kg-1 para 226Ra e 99,48 Bq kg-1 para 228Ra. Para 226Ra, os níveis não variaram significativamente com o tamanho das nozes, embora tais diferenças foram observadas para 224Ra e 228Ra. Não houve associação estatisticamente significativa entre o nível de selênio e a atividade de radionuclídeos, no entanto, houve correlação entre os radionuclídeos. Não foi encontrado aflatoxinas acima do limite de quantifcação.

RESUMEN

The aim of this study was to examine the beryllium-7 behavior in the soil. Natural variability of beryllium-7 concentration was calculated to be about 23 percent (relative standard deviation), and the depth distribution could be approximated by an exponential decay in bare soil, with an average penetration depth in the soil about 1 cm. The nuclide was not found below 2 cm depth, which confirmed its utilization to infer the erosion processes as a tracer of soil surface. The maximum beryllium-7 concentration in the analyzed period was about 40 Bq.kg-1.


Berílio-7 é um radionuclídeo cosmogênico, com meia-vida de 53 dias, produzido pelo processo de espalação de átomos de oxigênio e nitrogênio dentro da troposfera e estratosfera. Após sua produção, este é transportado até a superfície terrestre pela deposição úmida e seca. A precipitação seca contribui somente com 3-8 por cento do inventário total. Medidas de berílio-7 no solo podem serem usadas para indicar movimento de solo da camada superficial e este estudo objetiva examinar o comportamento de berílio-7 no solo. Variabilidade natural do inventário de berílio-7 é em torno de 23 por cento (desvio padrão relativo). A distribuição em profundidade de berílio-7 pode ser aproximada por uma função exponencial no solo nu, com uma profundidade média de distribuição no solo em torno de 1 cm. O berílio-7 não foi encontrado abaixo da profundidade de 2 cm para o tipo de solo estudado, o que confirma sua utilização para avaliar processo de erosão superficial como um traçador de solo superficial. A concentração máxima de berílio-7 no período analisado é em torno de 40 Bq.kg-1.

RESUMEN

The aim of this work was to assess the soil erosion process in native forest by the 137Cs methodology. The mass balance model was applied to assess the rates of soil loss in three native forests around of Londrina city, Paraná, Brazil. 137Cs distribution depth was of exponential type for the three forests and 137Cs inventory was 241 Bq m-2 for Mata 1, 338 Bq m-2 for Mata 2 and 325 Bq m-2 for Mata UEL. The soil loss value calculated for three native forests was: 6,684 kg ha-1 yr-1 for Mata 1, 1,788 kg ha-1 yr-1 for Mata 2 and 4,524 kg ha-1 yr-1 for Mata UEL.


O processo de erosão de solo em floresta nativa tem sido pouco investigado. Como a metodologia do césio-137 dá resultados tanto de taxas de erosão de solo como a bioturbação no perfil de solo, ele tem sido usado para avaliar o processo de erosão de solo nestes ecossistemas. O modelo de balanço de massa foi aplicado para avaliar as taxas de perdas de solo em três florestas nativas na região de Londrina, Paraná, Brasil. A distribuição em profundidade do césio-137 para as três florestas é do tipo exponencial. RESUMEN

O césio-137 introduzido no meio ambiente pelos testes nucleares de superfície é rápido e fortemente adsorvido às partículas finas do solo. Este pode ser usado como um marcador para determinação da redistribuição do solo. Esta metodologia tem sido muito usada nos países desenvolvidos, porém ainda muito pouco difundida no Brasil. O objetivo deste estudo foi a aplicação desta metodologia para determinação de perdas e ganhos de solo em uma bacia no município de Cambe, Paraná. Esta bacia está localizada ao redor das coordenadas 23º16' S e 51º17' O e o solo cultivado com diferentes culturas. Uma amostragem em transectos foi utilizada para avaliar a influência dos diferentes cultivos na erosão do solo. Um ponto em área de floresta foi amostrado e analisado para determinar o inventário de referência de césio-137 depositado pelo "fallout". O valor médio do inventário de referência foi de 292 Bq m-2. O inventário de césio-137 das amostras dos transectos variou de 80 Bq m-2 até 403 Bq m-2. Os pontos amostrados em pastagem e em cultura de soja apresentaram perdas de solo, e os pontos amostrados em plantação de café não apresentaram nem perda nem ganho de solo.
